TFF灌注细胞培养评估
单克隆抗体(mAb)是生物制药领域的主要研究对象之一。有几种类型的细胞培养系统,如补料分批和灌流培养。虽然传统的批次系统在当前的培养工艺中被广泛接受,但基于灌流细胞培养系统为长期大大增加细胞密度和生产力提供了潜力。中空纤维在灌流培养中作为细胞分离装置被广泛接受。我们对在TFF(切向流过滤)系统中灌流细胞培养的性能进行了评估。Test Conditions:
Cell culture: Antibody-producing Chinese hamster ovary (CHO) cells were cultivated in serum free media.
Cell density was maintained at 50-70 ×106 cells/mL by bleeding.
Medium exchange rate: 0.75-2.5 vessel/day
Permeate flux: 0.24-0.80 L/m2/h (LMH)
Bioreactor working volume was adjusted by membrane area.
Flow rate (L/min) was adjusted so that the shear rate inside of hollow fiber became 1500 s-1.
